If a local merchant in my area is selling an item can I buy a large amount from him and re-sell it on my website for a profit without his permission?

If you buy something, you can resell it.

Permission isn't needed to dispose of what is yours.

Army Judge is right. There is something called the First Sale Doctrine which, in most circumstances, allows you to resell what you bought, e.g. a painting. There are additional aspects of this rule but it may cover your concerns, whatever they may be - assuming a contract may not have explicitly restricted your sale.
