I bought a used 2005 Kia Optima. It currently has 50,00 miles on it. I took it into the shop because it was acting up and they told me I needed a new transmission and it was covered under warranty. Now they are telling me my warranty if voided because I didn't get my transmission service done at 30,000 miles. The owners manual says that for level one maintenance that I should do that at 100,000 miles. Only to do it at 30,000 if you drive under severe conditions. None of those conditions applied to me. What can I do?

Show them what you see on the booklet and prove to them that you have kept up with reasonable maintenance for your car (oil changes, tire pressure checks and the like). Unless they can prove to you otherwise, you are covered. If they refuse, tell them you will report the dealership to the local dealers association.
